Solid-phase peptide synthesis: the method behind research-grade material

Published chemistry literature describes how peptides are assembled stepwise on a solid support, and why coupling efficiency and purification strategy determine the quality of the finished research material.

Solid-phase peptide synthesis (SPPS) assembles a peptide chain one protected amino acid at a time on an insoluble resin. Published reviews describe repeated cycles of deprotection and coupling, followed by cleavage from the resin and removal of side-chain protecting groups.

Researchers have examined how incomplete coupling steps generate deletion sequences, and how oxidation or racemisation during synthesis produces closely related impurities that must be resolved chromatographically.

Preparative HPLC is the purification step most often reported in the literature, followed by lyophilisation to yield a stable powder for laboratory storage.
