What the field investigates
This area is heavily biochemical. NAD and its precursors are studied as substrates and cofactors: assays measure intracellular NAD and NADH pools, the activity of NAD-consuming enzymes such as sirtuins and PARPs, and how metabolic flux shifts when precursor availability changes.
Mitochondrial research uses respirometry to measure oxygen consumption, membrane-potential dyes to measure coupling, and lipidomic or spectroscopic methods to characterise interactions with mitochondrial membrane lipids such as cardiolipin.
